The LifeAssays® System

The LifeAssays® Reader offers a new alternative for POC testing utilizing the unique and patented magnet immuno assay (MIA) technique. The system composes of two parts:wet chemical reagents in the assays vial where the immunoreactions take place, and an analytical instrument, the LifeAssays Reader, for quantification and measuring. The two are connected with a disposable algorithm chip the “one computer chip” with all analytic specific data, as well as a self-executable calculation algorithm. The “one computer chip” is the LifeAssays® POC system’s key and locks the LifeAssays® Reader to the LifeAssays® reagents. The chip is inserted into the reader when a new reagent kit is opened and remains in the reader until the last reagent vial in the kit is disposed of. Upgrades are easily provided with each new reagent kit.

Performing a test using the LifeAssays® System is easy and convenient. Insert the reagent vial into the instrument and the LifeAssays® Reader will automatically perform the measurement and display the result after the completed reaction.

Description

The system composes of two parts:wet chemical reagents in the assays vial where the immunoreactions take place, and an analytical instrument, the VetReader, for quantification and measuring. The reagent is a two-site heterogeneous immunoassay, with a solid phase consisting of silica microparticles coated with target-specific antibodies. This is used as the solid phase. See figure 1.

The liquid phase consists of a sample buffer with magnetic nanoparticles which has also been coated with target-specific antibodies. These magnetic particles are colloidal, which means that they are small enough to stay in the solution, thus they do not sediment. The magnetic particles are used as the detector. See figure 2.

When the sample containing the target biological substance, i.e. the "marker", is placed into the reagent vial and the contents are mixed, the targeted marker will be captured by the target-specific antibodies on the silica surface. Target-specific antibodies on the magnetic nanoparticles will detect the marker bound to the silica surfaces in a sandwich-binding format. An immune-complex is formed. See figure 3.

The reagent vial is placed in the LifeAssays® VetReader and the instrument waits for the reaction to be completed before measuring the test result. During this time, the larger and heavier silica particles will sediment through normal gravity to the bottom of the reagent vial, forming a solid phase pellet. This pellet will now consist of both silica particles and a immuno complex of silica-coated-magnetic particles. The pellet will contain magnetic particles “fished” out from the fluid and will possess magnetic properties. See figure 4.

These magnetic properties are measured quantitatively with the internal coil in the LifeAssays® VetReader detecting the inductance. The change in inductance is proportional to the amount of magnetic material present in the pellet, and in turn shows how much of the selected marker that has been bound in the silica-coated-magnetic particle complex.

The internal coil measures the inductance 5 times by pushing the reagents vial in and out of the instrument,creating a “zero” value to use as a comparison each time the vial is lifted out of the instrument. An average raw signal value is created and sent to the “one computer chip” where the calculation takes place. The chip contains all analytic specific data and the self-executable calculation algorithm to calculate the test result in mg/L and displayed on the LifeAssays® VetReader.

This unique patented system has a number of advantages:

New tests

By changing the antibodies on the phases (silica and magnetic nanoparticles) a wide range of potential markers can be targeted. It is the properties of the target-specific antibodies that decide the specificity and sensitivity, and a wide of range of tests can be developed and run based on this system.

No instrument update needed

All information is stored on the “one computer chip” so the instrument does not need to be updated with new markers or software; the chip will make sure that the instrument is always running with the latest updates and with the correct batch information.

Quantitative system

The magnetic immunoassay technique in combination with the “one computer chip” allows for quantitative and objective measurements of biological markers. The system delivers numeric values for diagnosis and monitoring of patients.

Ideal point-of-care system

The system contains no optics.The tubing unit has a small footprint and weighs less than 1 kg, which makes it an ideal candidate for point-of-care settings. All updates are delivered with the “one computer chip” and require minimal maintenance and service. The unique technology is ideal for complicated sample matrices where, for instance, light scattering disturbance is an issue.

Research & Development

Human Pipeline

Over the years, LifeAssays has applied the unique platform for a number of potential diagnostic markers. Proof-of-principal work has been done on microalbumin in urine (u-Albumin), Cystatin C and Apo E.

More extensive work has been done on normal range CRP and high sensitive CRP. The normal CRP was CE-marked in 2011 and is currently undergoing further external evaluation and high sensitivity will follow during 2012.
